Formulation development and evaluation of flavoxate HCl gastroretentive floating tablets:(TJPS-2020-0267)
Abstract
Purpose: The objective of current study is to develop gastro retentive formulation for Flavoxate.HCl. HCl using polymers of natural and semisynthetic origin in appropriate composition. Flavoxate.HCl. HCl, a smooth muscle relaxant. Mainly used for treating urinary incontinence, urgency of urination. Methods: Floating tablets of Flavoxate.HCl. HCl were prepared using variable amounts of HPMCK100M, Lannea coromandelica gum (LCG) with effervescent mixtures as per 32 factorial design by direct compression technique. Amount of release modifiers need to get sustained drug release was labeled as factors. On other hand time taken for drug dissolution were labeled as Responses (t10%,t50%,t75%,t90%,). Results and Discussion: 9 formulations were obtained as per design and formulated and evaluated for quality control parameters. From the obtained results clears that all formulations passes the compendial limits. Data obtained from the disoolution study fitted well to kinetic modeling, kinetic parameters were determined. Polynomial equations were derived for Responses and checked for validity. Conclusion: FX5 composed of 40 milligrams of HPMCK100M & 40 milligrams of LCG, is the best formulation showing similarity f2=84.65, f1= 4.3 with the marketed product (URISPAS). Formulation FX5 follow first order, whereas release mechanism found to be nonfickian type (n= 0.76).
